Integrative single cell genomic analysis of mesenchymal stem cells
Mesenchymal stem cells (MSCs), which can be isolated from adult bones and teeth, are a major focus of regenerative medicine due to their versatile applicability. In this project, we will identify the mechanisms governing maintenance and differentiation of these elusive somatic stem cells by integrative single cell transcriptomic and epigenomic analysis. We will use a lineage-marking approach in a mouse model to isolate cells of interest and perform single cell RNA-seq and single cell ATAC-seq. Single cell genomic data of cells freshly isolated from bones, as well as cells clonally expanded in cultured conditions will be used for this project. We will then computationally integrate these datasets to identify corresponding cell types and investigate the epigenetic regulation of gene expression in mesenchymal stem cells.
